No, you can run only one JACK instance. But you can use the ~/.asoundrc file to agglomerate several hardware sound cards into one virtual card. In that case, it will work fine if one of the card can provide a world clock for the other cards. For that you will need professional sound cards or modify their hardware to send the clock of one card to the others. If you don't have that hardware synchronisation between the cards, their internal clocks will vary with time and you will get xruns or worst.

JACK can see only one card, but ALSA will still see the other cards. I use the ALSA "jack" plugin in my .asoundrc to send and receive the audio from the ALSA only softwares, and it work very well. It is some example here: http://proaudio.tuxfamily.org/wiki/index.php?title=DAW_Digital_Audio_Workstation#Using_the_ALSA_JACK_PCM_plugin

AFAIK, I just moved to http://proaudio.tuxfamily.org/wiki/index.php?title=DAW_Digital_Audio_Workstation#Using_an_ALSA_Loopback_device_and_zita-ajbridge
Took a couple of hours of fiddling to achieve, e.g. https://forums.gentoo.org/viewtopic-t-1093196.html.
Thks 4 ur attention, interest & support.
