May be you saw that, vbox forum suggests that some links should be fixed:

yes i seen that, i've added a line to my fstab as well concerning the usb file list.
it seams like it works and then if i do a reboot of the system it stops working.
it hit and miss. and yes i have auto load modual set. i have noticed when it fails i need to rebuild the modual and then do a reboot and it works. if i just do a reboot it continues to fail.

After years of using VirtualBox I just received this same error:

------
Failed to create the VirtualBox COM object.
The application will now terminate.

Callee RC:
NS_ERROR_FACTORY_NOT_REGISTERED
-----

I am using virtualbox 4.3.16.

I could run VirtualBox as root but no longer as an unprivileged user.

I fixed the problem by re-emerging all four virtualbox packages.

if anyone else has this problem, you may have tried to install virtualbox-bin then switched to virtualbox (non-bin)

to fix this problem, you need to fix the $VBOX_APP_HOME environment variable from

Code:
$ echo $VBOX_APP_HOME
/opt/VirtualBox

which no longer exists, as this error indicates, and change it to

Code:
export VBOX_APP_HOME="/usr/lib64/virtualbox/"

re-emerging did not fix for me, but above did work
