Yes, though AFAIK no-multilib profiles disable it by definition.
Make sure you go with a - let's call it - regular, hybrid install.

Yeah, I'm running multilib.
I ask because when I run

winetricks dotnet35

I get

This package does not work on a 64-bit installation. You must use a prefix made with WINEARCH=win32.

You'd think if wine had wow64 it would have no problem running 32 bit packages.

you'll could answer yourself with logic:
- wow64 works in wine64
- dotnet35 doesn't work with a wine64
--> wow64 doesn't need dotnet35

Maybe it is a good idea to first question: What is WoW64?

In short, it is a layer to make win32 applications work on win64.

So you only need that if you have to run 32 bit applications in a 64bit wine.

That leads to the next question: Do you really need a 64bit wine? Are you using any application that are 64bit-only and/or benefit greatly to be run in 64bit mode? And if so, why not simply creating a new 32bit prefix?

Wine on Gentoo does not support WoW64 like described on the page you linked.
