I have been having this problem over a year now and avoiding to come here with things that were caused by some people who marked stable packages too soon and or forgot their dependencies matter too.
Nothing was compiling in 3 gentoos and although i was able to fix 2 almost without knowing how i did it; the 3rd one is getting chaotic even tho it is the same hardware and almost the same build as the one i have a problem with.

Nothing compiles and when it not one thing it is the other until it gets redundant and i down see how to fix it.
udev fails due to autoconf/automake, if i fix autoconf/automake, fails due to dev-util/intltool being too old, removing it solves nothing and my other systems have the same packages working. When it is not this its because /usr/bin/glib-mkenums: /usr/local/bin/perl: bad interpreter: No such file or directory but removing, re-instaling, masking, upgrading, downgrading leads to the same, perl-cleaner fails until something else gets fixed; which never gets fixed, python-updater goes south too, removing packages and keep on removing has not solved anything so far and the list goes on.

I don't even know where to start but right now i need to solve this one:

I have re-installed de-lang/perl as well as upgraded glibc and recompiled and i cannot compile packages that need this and i am tired to browse pages for solutions that lead to bugs.

Looks weird. /usr/bin/glib-mkenums is part of glib. On my system it uses /usr/bin/perl not /usr/local/bin/perl. Can it be that when you've installed dev-libs/glib you have local version of perl installed in /usr/local/bin? Then possibly it was picked up during installation but then later you have removed local perl version.

I would recommend reinstalling glib to see if this would resolve the issue.

Okay, so all the previous evidence is destroyed. That solves that, I guess. ;)

The "configure: error: C compiler cannot create executables" message is typically caused by a syntax error in your CFLAGS variable in make.conf. Could you post you make.conf, please?

It is currently compiling with.

There was a mistake there that had -O2 -pipe maybe the "O" was a zero.

When I do 'ls /usr/local', I see these four folders:

bin
lib
lib64
sbin

So, is something awry with my directory in this regard?

it is normal to have these directories. If there is anything in them then you better know what you are doing ...

Jaglover, you are kidding me, right? :) Of course they're not empty. Why would they be empty? On the other hand, of couse I have no clue what I am doing!

No kidding. Stuff that goes to /usr/local is installed by hand and it is a great way to check out some unstable software without removing stable version, for instance. However, it is before /usr on your $PATH, meaning anything in there can be picked up by .configure and screw up your legit emerge builds.

Seems i'm too late, but rebuilding package / removing & reinstall package... while having ccache enable is certainly not helping you...

And if two of your hosts are ok with a package, it would had been faster and easier to build binary package of one of those host to install it on the problematic host.

In my case nothing was installed without portage. Whenever happened was with portage. Probably some "gremlin maintainer" of some package.
I have no explanation for this: /usr/local/bin/perl

I am now reinstalling my system.

If that came truly from portage, and not from some crazy overlay you installed, then it should be reported and fixed.

I guess your system is gone by now. But, if it isn't, could you please show us the output for these commands:

media-libs/mesa-9.2.5 also failed with -j3 but compiled with -j1
Re-installed proved to be a faster and less complicated option to take.
