I have 2 machines, one I'm just installing gentoo on. When you try to search I get => /usr/bin/locate: /var/spool/locate/locatedb: No such file or directory

This error occurs on both, one install is from 3-4 months ago (updated), the other is a new install this weekend. I probably wouldn't have found this error if it were'nt for some other errors/bugs I encountered installing gentoo this weekend.

Any ideas/fixes? Or should this be a gnome bug.

Type "updatedb" in console.

That worked :) . I'll have to do that for the other one. Does that mean I should updatedb frequently for it to know what's where?

Yep. If you have a crond installed, root's crontab isn't a bad place for it.
